It first opened its door for business in the year 2014 at Jurong East. Sunshine Supermarkets primary aim is to serve the customer. Keeping existing customers happy is important, as they are more likely to return. This is more cost effective for the business than acquiring new ones. Sunshine Supermarkets original product range of grocery and general merchandise has diversified to include home appliances, electrical goods as well as telephone equipment. In the past two year Sunshine Supermarkets growth has been massive mainly due to its excellent customer service and quality of products. Sunshine Supermarkets market expansion strategy is to penetrate further in the Singapore market by opening the second store in Punggol. Sunshine Supermarket would be recruiting 10 more retails staffs, three for the current branch and seven for the new branch. The new recruits would be foreign nationals with no prior retail experience. They would have to be trained in customer service, merchandising, stock taking and other functions. All employees have a varied style of learning. Three retails staffs who have been exemplary employees in the first branch will be promoted to the role of Store Manager, Purchase Manager and Supervisor respectively. It is important to understand Kirkpatricks Four Levels of Training Evaluation is just a method of evaluate. Identifying how to use will vary case to case basis. The Chemistry of Knowledge Hippeas thought he had all the answers. â€Å"I have never found any man who was my superior in anything,† he boasted. Then he meets Socrates. Though he had made thousands of public speeches about virtue, a dialogue with the wisest of Athenians leads Hippeas to confess that he â€Å"cannot even say what [virtue] is† (Hippeas 70). Lesser Hippeas discredits Hippeas but offers little more than a negative definition of knowledge. Meno, Phaedo, and the Republic provide a more comprehensive discussion of the definition, the good and the teaching of knowledge. The following pages will explore Plato’s theory of knowledge and will conclude with an examination of organic chemistry at Swarthmore College. According to Plato, knowledge requires the reasoned understanding of essences. To know the qualities something possesses, one must grasp â€Å"what something is† (Meno 60). Knowledge entails an active understanding capable of weathering the rigors of a probing discussion. In Lesser Hippeas, Socrates uncovers the knowledge deficit underlying Hippeas’ opinions and skills. Plato makes a clear distinction between Hippeas’ mastery of facts and the possession of true knowledge: â€Å"I certainly do not think I am guessing that right opinion is a different thing from knowledge† (90). Facts and opinions require little more than memorization and regurgitation. Acquiring knowledge, on the other hand, can only be done â€Å"with an effort† (Republic 776). Plato’s Cave analogy in the Republic likens the path to knowledge to the transition from a dark cave to a sunny hilltop. Plato allegorizes the â€Å"ascent of the soul† (776) from a world of shadows, reflections and half-truths into the â€Å"world of knowledge† (776). For me, perhaps the most disconcerting thing about organic chemistry is the rapidity with which my memory of it has faded. Some three months removed from a final exam, I recall virtually nothing. Professors need empirical measurements of student progress in finite periods of time, even when it is clear that cursory thought emasculates knowledge. Jewish merchants had become very competitive which supposedly put some fear on others. Consequently, in 945, the Venetian government ordered its ship owners not to carry Jewish passengers .   Ã‚  Ã‚  Ã‚  Ã‚  Jewish merchants traded in amber, textiles, hides, arms, spices, precious stones, and other luxury articles . Their clientele consisted mainly of royal and ducal courts and the aristocracy, both secular and clerical . In the cities re-conquered from the Muslims in Spain, Jews played a decisive role in the revival of commerce and industry, and especially in the production and merchandising of clothing . Various speculations and educated guesses place our total depletion of crude oil within the next 50 years and there is a general consensus between environmentalists that we steer toward a hydrogen transportation system given the projected work and nonexistent carbon dioxide emissions (Environmental Technologies class lecture, Santa Clara University). However many barriers stand in the way of attaining such a goal, most of which pertaining to the conservative nature of society. Society works in intermediary changes, a series of steps that blend rather than abandon one method of living for another. The option 22 states have chosen, is ethanol a combustive liquid fuel that could progressively move the United States toward a clean and su stainable environment and economy. Ethanol is an alcohol molecule consisting of two carbon molecules and six hydrogen molecules. Ethanol production in the United States results as a chemical byproduct from our most abundant food stock†¦ corn. Through four well known and commonly practiced chemical processes, Hydrolysis, Fermentation, Distillation and Dehydration. Major similarities between FASB and IASB: Same organizational structure FASB and IASB are each belongs to an organization that has four main bodies. Also, the internal structure and responsibility assignment of these organizations are basically the same. FASB and IASB are the standard setting board of the organizations. Other three parts of the organizations are there to support the standard setting process, such as selecting board members and deal with emergency issues (Kieso, et al. , p7-9). In addition, both boards have members from varies industries including accounting, finance, business and academia (Virginia Law & Business Review, fall 2008, p283). Private entities Both boards are private entities that have been assigned the role and responsibility of developing and deploying Accounting Standards (Virginia Law & Business Review, fall 2008, p284). By private, it means the standard setting process is not affected by politics. However, since the governments are also relying on standard set by these private institutions, both FASB and IASB are sensitive to politics to varies degree. Similar procedure of making rules Despite any internal and external influences, the procedure of drafting and publishing Generally Accepted Accounting Principles (GAAP) and International Financial Reporting Standards (IFRS) are similar. Generally, the process is as follows: discuss whether to add new project to agenda mostly based on recommendations from investors; plan for the project; draft the standard; vote and publish the standard once it is approved(FASB: Financial Accounting Standards Board, n. d). However, since each board has different members and they are setting rules based on its own investor’s concern, also by other external influences, they produce distinct rules. Differentiations between FASB and IASB: Scope of operations The jurisdiction of the IASB could be the whole world of commerce and industry, include EU nations and states, the latter is confined mainly to the United States and its trade partners (Virginia Law & Business Review, fall 2008, p303). Because of this, the citizenship of the trustees has become a problem. Since many of your clients are Fortune 300 companies who would find FASB more in harmony with their preparation and presentation of periodic accounting statements and records. FASB is also in line with most widely accepted accounting norms and practices in the US. With less external influence, more freedom in standard setting and sufficient funds, I believe the FASB standard setting process is more trustworthy.

Society labeled these people to be against the norms or conformities, which in return allowed the patients to feel inferior and out of place. This novel stresses the fact that each person should not be forced into a corner; they should be given rights to live regular lives with other people. Society should not force inferiority complexes on these patients. The fact that most of the patients were voluntary helps prove this point. It shows how society forced them into a completely different and inhumane lifestyle. Another novel that joins this rebellion against society in relation to insanity is The Bell Jar by Sylvia Plath. In this novel, Plath interprets insanity in her own way trying to prove practically the same point as Kesey. Plath’s use of description, metaphors, and characterization help provide her main point of allowing insanity to merge with reality. The Bell Jar is a novel equipped with descriptions that allow the reader direct access to the main character’s mind. Plath uses an abundant amount of physical descriptions such as the description of Esther’s surroundings at all times to help give examples of how society has forced this woman to think. Esther’s thoughts on life, death, and the world all seem to be reasonable and justifiable thoughts. She is capable of convincing the reader that those thoughts are not insane. Through the use of descriptions, Plath was able to highlight the unjust life of a 1950s woman. Plath also uses metaphors to highlight the suppression made by society on the women. The title of the book is the major metaphor that best represents Plath’s idea on society’s conformities. The entire novel revolves around the idea of the bell jar and this jar represents how society analyzes and reduces the contents provided in the jar. The jar represents insanity. Esther feels secluded and isolated from the real world when she is labeled as mad. She feels like she is an airless jar that ruins her perspective of what the real world is. It signifies a buffer that ends the connection between Esther and the real world4. These literary features were all used to highlight Esther’s alienation from the real world. It shows how a young woman from the 1950s was forced to act. Esther wanted to pursue a writing career and is supported completely, but her thoughts begin to change when the fact that she cannot merge her career with being a mother come to place. Esther becomes depressed and her thoughts begin to change on the world. These individual thoughts begin to accumulate leading to actions that are condemned upon by society. Society expects a lot from Esther such as the idea of her virginity. Esther rebels against the conventional role of virginity with women at that time by embarking on a sexual experience. Esther did not become insane because she believed against the norms of society but rather insanity fell on her. The treatments in both novels are similar in many aspects. At first, the hospitals provide healthy conversations between the patient and a professional psychiatrist. In both novels, the sense of talking is important because a lot of information is released about society and what they think of it. Another treatment usually done after talk therapy is electroshock therapy. Electroshock therapy was created in 1936 in order to help patients clear their mind5. As years passed, this treatment began to evolve which therefore led to the change of its purpose. In One Flew Over the Cuckoo’s Nest, ECT was used as a form of punishment. Patients were punished for doing anything out of the ordinary. In The Bell Jar, Esther Greenwood undergoes several electroshock therapy sessions to clear her mind. She continuously states how painful the therapy is and tries to refuse treatment. Her recollections of these treatments show the inhumanity in medical treatments. Another treatment that falls into the controversial category is Lobotomy. It is shown insignificantly in The Bell Jar, one patient briefly converses about it. On the other hand, Lobotomy has a major impact in One Flew Over the Cuckoo’s Nest. The main character undergoes a lobotomy at the end of the novel. This act completely criticizes the ways of society in relation to medical treatments. Many forms of medical treatments on insanity exist but whether they provide a positive outcome is the main question. Society and mental illness are very closely related in the sense that society creates the separation between sanity and insanity. That separation is miniscule and changes constantly over time. Both of these novels emphasize isolation, suppression, and seclusion forced by society. Society forces these on the ideas that are condemned or not wanted. These ideas should not force inhumane actions but rather welcome ideas as an advantage to a better society, a more open society. Prejudice In Organizations Essay

?Question 2: Prejudice can be hurtful and destructive – discuss how you can personally reduce prejudice in your workplace – please provide an example. Prejudice can be described as a word which is derived from the Latin word Prae Judicium meaning ‘to try in advance’ (Clawson et al: 1990). Prejudice happens when we pre-judge individuals on first encounter about their character or appeal. Most individuals who are prejudiced are usually rigid in their prejudices and their beliefs are unsubstantiated. Prejudice can create serious tension in an organization because it has the potential to strain interpersonal relationships in a workplace. People can practice prejudices in various forms. Some of the areas of potential prejudice could be gender, race, age, religion, sexual orientation, ethnicity and physical disabilities. Prejudices can be systematically dealt with in the workplace. I work for one of the United Nations (UN) agencies. The UN has noted and is mindful of potential prejudices that can exist in the workplace, especially that the workplace is made up of employees from various countries, backgrounds, ethnic groupings, varying religious beliefs (including atheists), sexual orientation, races and as an organization, The organisation has a very diverse workforce. This diversity has not been ignored. The UN has taken deliberate steps to introduce workplace policies that encourage tolerance amongst employees and in turn, tolerance in the communities in which we serve. The policies are part of the orientation package for every new employee who joins the UN. There are Executive Directives (EXDs) that are issued and reviewed on an ongoing basis. In the current directive, disagreement between a staff member and supervisor is not considered as prejudice. The policies are clear on the definition of prejudices and also on the consequences of perpetuating the vice. If I were to be found in a work environment where prejudices are rife, I would first of all admit that prejudices are real and we all have some form of prejudices. The first step to deal with a problem is to admit that you have one. You can only begin to work on one’s prejudices once they realize they have them, and they can begin to work more carefully with the prejudices of others without anger and force (Clawson et al: 1990). It’s a realization that we all have them that helps us to deal with them. Members of staff in an organization, must be allowed to deal with perceived prejudices. It is not advisable to pretend that the problem does not exist as it becomes difficult to deal with a hidden or unperceived problem. Once the problem has been identified, then comes the process of working on your own as well as the prejudices of others in the organisation. This can be achieved through redirecting prejudiced statements by colleagues towards functional discrimination e. g. instead of lamenting how a workmate or subordinate is not achieving her objectives because she is a woman, this can be countered by a statement that shows how previously, a male colleague had also failed in a similar position to show that, gender has nothing to do with functioning in a position. For prejudices which are merely based on ignorance like â€Å"all Muslims are terrorists†, it would be helpful to team up Muslim staff members and some of the staff members who are holding on to this kind of prejudice. That way, it allows them to interact at a personal level and get the truth about Islam. If this doesn’t work, then interactions between such colleagues should either be kept to the minimum or topics of discussion should stir away from sensitive issues. I would also learn to listen to others with an open mind, not listening with an intention to respond as this is likely to attract judgmental behaviour. In a multi ethnic organisation, learning about the cultures of other countries helps us be more tolerant. People are more likely to react in a certain way because of the environments they have been brought up in. Prejudices will always exist in organisations, it would therefore helpful to encourage tolerance among employees, this can be achieved through deliberate company policy, sensitisation and clear consequences for behaviour promotes prejudice.

If staff members did not carry out frequent bar, floor and toilet checks then both employees and customers will be at risk of a potential risk. This could be slipping on a spilt drink or the toilets being unhygienic. Staff need to constantly make the bar, floor and toilet as clean and safe as possible. This includes wiping spillages on the bar, collecting glasses and plates and sweeping and moping. Kitchen Staff: Kitchen staff have a very high responsibility and role within the company regarding health and safety, as they are handling food. Kitchen staff have to ensure that their hands are constantly being washed, the kitchen is clean and food is in date. If kitchen staff did not have a responsibility within health and safety then customers could potentially be food poisoned, as well.

The next step in assessment is to determine the etiology and risk factors of the wound. Age, malnutrition, collagen disease and drugs like steroids inhibit wound healing (Kunimoto, 2001). After medical history, physical examination of the wound must be undertaken. Physical examination gives valuable information especially in the elderly wh o cannot recall history details. One of the aims of physical examination is to differentiate between venous leg ulcers and non-venous leg ulcers. Venous leg ulcers occur in the distal medial third of the lower extremity and they are shallow without punched margins. The amount of granulation tissue and its color must also be assessed. Nurses are often confused here. Differences in documentation of the quantity and color of granulation tissue by the nurses have been reported (Frank-Stromborg et al, 2001). Decreased granulation tissue or bluish hue color of this tissue are indicators of bacterial infection. The drainage from the wound must be examined as to whether it is serous, purulent or serosanguinous. There is evidence that there is no homogeneity among nurses and other health professionals in ascertaining the type of secretion from wound (Stremitzer et al, 2007) and this aspect plays a very important role in wound evaluation and diagnosis. Purulent discharge is an indicator of ba cterial infection. The length, depth and width of the ulcer must also be assessed. Surrounding skin examination gives a valuable clue to differentiate between venous and non-venous ulcers. Peripheral arterial circulation assessment is a must in ulcer evaluation. Activation Of T Lymphocytes Essay Different types of T lymphocytes exist and each type performs a specialized function.   Helper T cells, also known as TH cells, serve as messengers of the immune system.   When helper T cells are activated, these cells proliferate and produce cytokines, which are small proteins that control the extent of immune response that a biological organism will express.   Several types of cytokines exist and each cytokine triggers the maturation of helper T cells into specific subtypes such as helper T cell subtype 1, 2 and 17 (Balandina et al. , 2005). Another type of T lymphocyte is the cytotoxic T cell (CTL), which is also known as the TC cell.   Cytotoxic T cells are responsible for the destruction of cells that were infected by viruses, as well as for the annihilation of tumor cells.   These cells also play a major role in determining whether a cell is normally part of the system or a foreign cell.   Cytotoxic T cells are known to be the first in line to react and reject if an organ transplanted into a recipient is not compatible in terms of tissue compatibility. The cytotoxic T cells that are associated with tissue rejection are designated at CD8+ T cells, based on the idea that these cytotoxic T cells produce a specific CD8 glycoprotein that is displayed on its cell membrane.   These CD8 glycoproteins interact with helper T lymphocytes, of which may trigger its transformation into regulatory T cells.   The major role of regulatory T cells is to inhibit the onset of an autoimmune disorder within the system of the organism. Another type of T lymphocyte is the memory T cell which is produced as soon as an infection occurs in an individual.   These specific T cells continue to circulate in the blood system for several months even when the infection has been stopped.   Memory T cells further differentiate into effector T cells when they find the same antigen that was associated with the previous infection, hence the term memory is appropriate to apply to this type of function.   Two general types of memory T cells include the central and effector cells.  Ã‚   Memory T cells are known to carry either the CD4 or the CD8 glycoprotein on its plasma membrane (Wildin et al., 2001). Regulatory T cells are another type of T lymphocytes that play a crucial role in sustaining the tolerance of the immune system (Khattri et al., 2003).   These T cells were earlier called suppressor T cells and their primary role is to prevent immunity that is caused by other T cells during the later stages of an immune response (Fontenot et al., 2003).   Regulatory T cells are also responsible for inhibiting the immune reaction towards other cells of the body or self cells because this is important in maintaining the stability of the entire immunological system. Specific conditions result in the immune response towards self cells and this is usually observed when the thymus experiences difficulty in distinguishing self cells from non-self or foreign cells.   There are two major types of CD4+ regulatory T cells that have been characterized.   Naturally occurring regulatory T cells originate from the thymus and are sometimes referred to as CD4/CD25/ForP3 T regulatory cells. The other major type of CD4+ regulatory T cell is the adaptive T regulatory cell which is produced when an innate immune response occurs.   This type of cell is also called Tr1 or Th3 cell (Hori et al., 2003).   A distinguishing feature that facilitates is the differentiation of naturally occurring T regulatory cells from the rest of types of T lymphocytes is the occurrence of the FoxP3 molecule that is situated within the cytoplasm.   Research has shown that mutations incurred in the FOXP3 gene results in the inability of regulatory T cells to differentiate, which in turn influences the onset of the lethal condition of autoimmunity. Another type of T lymphocyte is the natural killer T cell which is serves as the communicator between the adaptive and the innate immune systems.   Different from the standard T lymphocyte that identifies protein antigens that are harbored by the major histocompatibility complex (MHC), the natural killer T lymphocytes distinguish glycolipid antigens that are shown by the protein molecule CD1d.   The natural killer T lymphocyte is then activated and subsequently acquires the ability to execute specific functions that are related to Th and Tc cells, including the secretion of cytokines and the expression of cytolytic molecules that are responsible for the destruction of foreign cells that have invaded the body. Gamma-delta T lymphocytes represent a small subtype of T cells that carry a unique receptor on their plasma membrane.   Most of the T lymphocytes carry a receptor that is comprised by two strings of alpha and beta glycoproteins.   In gamma-delta T lymphocytes, the receptor is composed of a gamma and a delta glycoprotein string.   Gamma-delta T lymphocytes are quite rare, comprising only approximately 5% of the entire T lymphocyte population but are found in high concentrations in the epithelial lining of the stomach, as part of population of intraepithelial lymphocytes. Protein antigens that trigger a response from gamma-delta T cells have still yet to be determined.   It is interesting to know that gamma-delta T lymphocytes are not exclusively assigned to carry major histocompatibility complex molecules and are actually capable of identifying entire proteins than simply perceiving short protein chains carried by the major histocompatibility complex molecules on lymphocytes that harbor antigens. There are particular gamma-delta T cells that have the capacity to identify a specific type of major histocompatibility complex molecules, specifically class IB.   In the human immune system, the Vgamma9/Vdelta2 T lymphocyte makes up the majority of the gamma-delta T lymphocyte population that is circulating in the bloodstream.   These specific T lymphocytes have the function of quickly reacting to a tiny metabolite that originates from microorganisms, known as the isopentenyl pyrophosphate precursor.   Another type of T lymphocyte is the autoaggressive T cell which is highly specialized based on its ability to secrete the CD40 protein molecule (Miura et al., 2004).   CD40 is generally linked to T lymphocytes that present fragments of an antigen and this protein molecule is commonly produced by a subtype of T helper lymphocytes. Th40 cells are present in all human beings but the levels of this specific type of cells quickly increase during conditions of autoimmunity.   In patients diagnosed with type 1 diabetes mellitus, the Th40 cells usually react to self-antigens.   In the case of patients diagnosed with non-autoimmune individuals, the Th40 lymphocytes do not react with these antigens.   An important function of the CD40 protein molecule on T lymphocytes is to trigger the recombinase proteins RAG1 and RAG2 in directing the receptor of a T lymphocyte. The receptor of a T lymphocyte serves as the route through which a T lymphocyte is able to identify a specific antigen.   It has been determined that the two recombinase proteins be produced exclusively in the thymus during the development of T lymphocytes.   It was, however, observed that the RAG proteins were secreted again by the T lymphocytes that were circulating in the bloodstream.   In addition, it was also observed that the CD40 protein associated with Th40 lymphocytes, resulting in the production of RAG proteins.   Subsequent to the expression of the RAG proteins, modifications in the receptor of the T cells take place, suggesting that the Th40 lymphocytes carry the ability to adapt for the entire duration of an individual’s life. The procedure of modifying the expression of the receptors of T cells that are circulating in the bloodstream has been names as TDR revision.   Research has reported that TCR revision influences the expansion of the repertoire of T lymphocytes, as well as generates T lymphocytes that are autoaggressive.   This observation therefore shows that the revision of the receptors of T cells is another method of T lymphocyte tolerance. THE DEVELOPMENT OF T LYMPHOCYTES IN THE THYMUS T lymphocytes are developed from the stem cells of the hematopoietic system which is located in the bone marrow.   These stem cells migrate from the bone marrow to the thymus, where they are expected to proliferate through the process of cell division and generate immature thymocytes.   Young thymocytes generally do not secrete any CD4 or CD8 proteins hence these cells have been described as CD4-CD8- cells. During their course of development, the young thymocytes acquire the capability of secreting CD4 and CD8 molecules and these thymocytes are now classified as CD4+CD8+ (Sakaguchi, 2000).   Further maturation of these cells involves the specific secretion of only one protein, either the CD4 or the CD8 molecule.   Once this has been determined in each thymocytes, these cells are then expelled by the thymus in order for them to circulate to the rest of the body.   Approximately 98% of the thymocytes produced by the thymus are expected to disintegrate during its maturation, resulting in only 2% of the thymocytes reaching maturity and categorically expelled by the thymus as immunocompetent T lymphocytes. Immature thymocytes that harbor the two types of antigens, CD4 and CD8, migrate into the core of the thymus in order to associate with antigens that are generated by the T lymphocyte.   These thymocytes interact with the major histocompatibility complex molecules that are present of the plasma membrane of the cells of the epithelium.   Only a specific fraction of thymocytes will be able to completely bind with the major histocompatibility complex molecules thus resulting in a signal that indicates the survival of the thymocyte. The rest of the thymocytes that were not able to achieve a complete association with the major histocompatibility complex molecules are thus classified as incapable of performing immune functions or even illicit an immune response.   This specific group of thymocytes thus undergoes the process of apoptosis, which involved the disintegration of the nucleus which eventually leads to cell death.   The cellular debris that results from the apoptotic pathway are enveloped and eaten by macrophages.   The entire process of differentiating immuno-competent from immuno-incompetent thymocytes based on the ability to illicit an immune reaction is known as positive selection. Thymocytes that endure positive selection travel towards the perimeter of the cortical and middle regions of the thymus. While situated in the middle region of the thymus, the thymocytes are presented with an antigen of its self in association with the major histocompatibility complex molecules on antigen-presenting cells (APCs), including the dendritic cells and macrophages.   Thymocytes that intensely associate with the antigen are sent a signal that induces their programmed death and the rest of the thymocytes that were first generated are trigger to disintegrate during this selection process in the thymus. A small fraction of the cells that survived is induced to develop regulatory T cells.   The other cells subsequently depart the thymus as differentiation naive T lymphocytes.   This mechanism is designated as negative selection, an essential process of immunological tolerance that controls the development of T lymphocytes that react to self cells and are able of to cause an autoimmune disease in the body of an individual (Shevach, 2000). Although the specific mechanisms of activation vary slightly between different types of T cells, the two-signal model in CD4+ T cells holds true for most. Activation of CD4+ T cells occurs through the engagement of both the T cell receptor and CD28 on the T cell by the Major histocompatibility complex peptide and B7 family members on the APC respectively. Both are required for production of an effective immune response; in the absence of CD28 co-stimulation, T cell receptor signalling alone results in anergy. The signalling pathways downstream from both CD28 and the T cell receptor involve many proteins. The first signal is provided by binding of the T cell receptor to a short peptide presented by the major histocompatibility complex (MHC) on another cell. This ensures that only a T cell with a TCR specific to that peptide is activated. The partner cell is usually a professional antigen presenting cell (APC), usually a dendritic cell in the case of naà ¯ve responses, although B cells and macrophages can be important APCs. The peptides presented to CD8+ T cells by MHC class I molecules are 8-9 amino acids in length; the peptides presented to CD4+ cells by MHC class II molecules are longer, as the ends of the binding cleft of the MHC class II molecule are open. The second signal comes from co-stimulation, in which surface receptors on the APC are induced by a relatively small number of stimuli, usually products of pathogens, but sometimes breakdown products of cells, such as necrotic-bodies or heat-shock proteins (Brunkow et al., 2001). The only co-stimulatory receptor expressed constitutively by naà ¯ve T cells is CD28, so co-stimulation for these cells comes from the CD80 and CD86 proteins on the APC. Other receptors are expressed upon activation of the T cell, such as OX40 and ICOS, but these largely depend upon CD28 for their expression. The second signal licenses the T cell to respond to an antigen. Without it, the T cell becomes anergic and it becomes more difficult for it to activate in future. This mechanism prevents inappropriate responses to self, as self-peptides will not usually be presented with suitable co-stimulation. The T cell receptor exists as a complex of several proteins. The actual T cell receptor is composed of two separate peptide chains which are produced from the independent T cell receptor alpha and beta (TCR? and TCR?) genes. The other proteins in the complex are the CD3 proteins; CD3 and CD3 heterodimers and most importantly a CD3? homodimer which has a total of six ITAM motifs. The ITAM motifs on the CD3? can be phosphorylated by Lck and in turn recruit ZAP-70. Lck and/or ZAP-70 can also phosphorylate the tyrosines on many other molecules, not least CD28, Trim, LAT and SLP-76, which allows the aggregation of signalling complexes around these proteins. Phosphorylated LAT recruits SLP-76 to the membrane, where it can then bring in PLC?, VAV1, Itk and potentially PI3K. Both PLC? and PI3K act on PI(4,5)P2 on the inner leaflet of the membrane to create the active intermediaries di-acyl glycerol (DAG), inositol-1,4,5-trisphosphate (IP3) and phosphatidlyinositol-3,4,5-trisphosphate (PIP3). DAG binds and activates some PKCs, most importantly in T cells PKC?, which is important for activating the transcription factors NF-?B and AP-1. IP3 is released from the membrane by PLC? and diffuses rapidly to activate receptors on the ER which induce the release of calcium. The released calcium then activates calcineurin, and calcineurin activates NFAT, which then translocates to the nucleus. NFAT is a transcription factor which activates the transcription of a pleiotropic set of genes, most notably IL-2, a cytokine which promotes long term proliferation of activated T cells. Overview of interactions between T cells and APCs T cells are a subset of lymphocytes that play a large role in the cell-mediated immune response of adaptive immunity. T lymphocytes are divided into 2 major classes distinguished by the expression of the cell surface molecules CD4 or CD8. CD8 cytotoxic T lymphocytes (CTLs) primarily destroy virus-infected cells whereasCD4 helper T lymphocytes are involved in activating B cells and macrophages. The response of both classes of T lymphocytes is dependent on interactions through the T cell receptor (TCR) and coreceptors (CD4 or CD8) with target APCs that present peptides in the context of MHC class I or class II molecules. MHC class I and class II molecules are structurally similar but differ in their source of antigenic peptide and in the transport mechanisms for their peptides. Class I MHC molecules present self peptides or viral products from the cytosol to CD8 T lymphocytes, whereas MHC class II molecules present peptides from pathogens that either reside or were endocytosed into intracellular compartments (Fontenot and Rudensky, 2005). T cells are activated on encountering antigen, which results in either lysis of target cells by CD8 T lymphocytes or recruitment of other effector cells by CD4 cells.   Modulation of the plasma membrane alters the T cell Ca2+ response.   A series of studies from showedthat unsaturated free fatty acids (FFAs) could inhibit specific aspects of cytotoxic T cell function by perturbing membranes.   Initially, it was shown that short-term exposure of murine allogeneic effector T cells to low levels of unsaturated FFAs (10 mol/L), including PUFAs, inhibited lysis of target APCs. The change in lysis of target cells was a direct consequence of the FFA added to the CTLs, because lysis could be inhibited by extracting the unsaturated FFA with bovine serum albumin before CTL-target conjugation. Specific aspects of T cell function inhibited by unsaturated lipids included the initial rise in intracellular [Ca2+] on conjugate formation, protein phosphorylation events and subsequent CTL esterase release.   On the other hand, release of inositol phosphates and binding to target cells were unaffected. Because the inhibition in CTL calcium release linearly correlated with the decrease in membrane acyl chain order induced by the presence of increasing unsaturation in the plasma membrane , it was hypothesized that modulation of membrane structure affected T cell Ca2+ signaling.
